Histological Analysis of an Alkaline Based Tissue Clearing Technique for Fetal Musculoskeletal Tissue

Tissue clearing techniques are frequently used in the observation and description of anatomical structures pathways without altering three-dimensional layout specimen. optical promotes preservation structures, which allows study internal anatomy its original position spatial interaction. Among these techniques, Potassium Hydroxide (KOH) maceration is one most widely used. However, histological changes tissue after KOH have yet to be fully understood. Our aim describe microscopical differences between macerated normal tissue. To better understand said changes, two human fetuses with a gestation period 16 28 weeks were cleared processed for analysis. Microtome slides fetusesâ lower limbs obtained stained Hematoxylin & Eosin, Periodic Acid Schiff (PAS), Massonâs trichrome purpose observing macromolecule composition Remarkable at level regarding cellular since diaphanized tissues showed predominance extracellular matrix composed collagen fibers absence nucleated Phospholipidâs saponification, nucleic acids degradation change on proteins structural properties main factors inducing clearing. At same time, molecular stability alkaline conditions specimen maintain shape process.
